The dilution factor formula

The dilution factor (DF) is the ratio of the final total volume to the sample volume you started with:

DF = V_final ÷ V_sample

where:

  • V_sample = volume of the aliquot taken from the original solution
  • V_final = total volume after adding diluent (sample + diluent)

The diluent you actually add is the difference:

V_diluent = V_final − V_sample

A dilution factor of 10 is the same as a 10× dilution or a 1:10 ratio, one part sample to nine parts diluent (not one part to ten). The volume units cancel, so any consistent unit works (µL, mL, L) as long as both inputs share it.

Worked example

You pipette 2 mL of a bacterial culture and bring it up to a final volume of 20 mL with broth.

  • DF = 20 ÷ 2 = 10 → a 10× dilution.
  • Diluent to add: 20 − 2 = 18 mL.
  • Ratio: 1:10 (1 part sample + 9 parts diluent).

So your measured count must be multiplied by 10 to recover the original concentration.

Common pitfalls

  • 1:10 ≠ 1 part in 10 parts diluent. A 1:10 dilution is 1 part sample + 9 parts diluent for a total of 10, the factor counts the final volume, not the diluent alone.
  • Mismatched units. Keep V_sample and V_final in the same unit; mixing µL with mL inflates or shrinks the factor ×1000.
  • Final volume must exceed the sample. If V_final ≤ V_sample you are not diluting; a factor ≤ 1 means concentrating, which adding diluent cannot do.
  • To recover the original value, multiply by the factor. A measured concentration from a 1:100 dilution is ×100 below the true value.
